本研究團隊以三角褐指藻 (Phaeodactylum tricornutum) 做為模式生物,成功利用三角褐指藻的鹼性磷解酶啟動子 (alkaline phosphatase promoter, 簡稱pPtAPase) 構築了一個能夠受到缺磷逆境的誘導,進而大量進行基因表現與重組蛋白製造的表現載體。相較於其它的矽藻表現載體pPha與pNR, 我們開發的pPtAP載體所表現出的EGFP外源性基因重組蛋白能夠高出13倍或17倍。此外,在基因轉殖矽藻細胞中重組蛋白質的數目高達總蛋白量的50%以上。
由於矽藻的重組蛋白表現系統的培養成本低廉,基本上只需要光照與海水營養鹽。矽藻具備高度後轉譯修飾能力(post-translation modification),能夠表現結構比較複雜的重組蛋白。可望將此系統廣泛應用在合成單株抗體、疫苗等醫療用重組蛋白方面。

Here, we identified a functional promoter of PtAPase of the alkaline phosphatase (pPtAPase), which is highly induced in Phaeodactylum tricornutum. By using it, we also developed a novel diatom expression vector (pPtAP), which can induce exogenous gene’s expression abundantly when cells are phosphorus starved. Compared with other diatom’s expression vectors, such as pPha and pNR, our pPtAP vector expressed 13- and 17-fold of EGFP exogenous recombinant proteins, respectively. Moreover, the abundance of recombinant proteins is as much as 50% of total protein in the transgenic diatom.
